abstract | The present invention provides a composition and method for reducing the expression of an intracellular target gene, comprising contacting the cell with a double-stranded nucleic acid (dsNA) isolated in an effective amount to reduce the expression of an intracellular target gene. The dsNA of the present invention has a deoxyribonucleotide pattern designed to indicate the dicer enzyme cleavage site in the dsNA molecule (in most embodiments, the pattern comprises one or more deoxyribonucleotide-deoxyribonucleotide base pairs) . The deoxyribonucleotides of the dsNA molecules of the invention are cleaved through the cleavage site to produce active siRNA agents that no longer contain a deoxyribonucleotide pattern (e. G., A deoxyribonucleotide-deoxyribonucleotide base pair) Lt; RTI ID = 0.0 > dsNA < / RTI > The DNA extended dicer substrate siRNA (DsiRNA) has proven to be an effective RNA inhibitor more than the corresponding double-stranded RNA-extended DsiRNA.
