titleOfInvention | Method for screening yeast strains with enhanced recombinant protein secretion using yeast variants having cell-wall defect |
abstract | The present invention relates to a method for screening a yeast strain that secretes an introduced target protein, a method for enhancing the secretion ability of a target protein, a method for producing a target protein, and a method for producing yeast mutants having improved secretion efficiency of Gas1 or its fusion protein. The yeast strain expressing the fusion protein in which the target protein produced by using the GAS1 deficient strain inhibited growth under the cell wall stress condition of the present invention is bound to the N-terminal of the Gasl protein is characterized by cell wall formation due to secretion of the fusion protein It is possible to effectively identify yeast strains with increased secretion ability by confirming the increase in growth ability, and thus can be usefully used for screening strains having improved secretion ability from a series of mutant libraries prepared by random mutagenesis for mass production of recombinant proteins. |
