abstract | The present invention relates to a soluble fragment of influenza virus RNA-dependent RNA polymerase subunit PB2 and variants thereof, and to a crystallized complex thereof comprising an RNA cap analogue. The present invention also relates to a computerized method of screening and designing compounds that interact with RNA cap binding pocket using the structural coordinates of the complex. In addition, the present invention preferably binds to a PB2 polypeptide fragment comprising an RNA cap binding pocket by using the PB2 polypeptide fragment in a high-speed setting, and preferably inhibits interaction with an RNA cap or an analogue thereof To a method for identifying a compound. The present invention also relates to a compound for treating a disease state due to a viral infection caused by a negative-sense single strand RNA virus, and a pharmaceutical composition comprising the identified compound. |
