abstract The present invention relates to ethynyl derivatives of formula (I) or pharmaceutically acceptable acid addition salts thereof, racemic mixtures, or the corresponding enantiomers and / or optical isomers and / or stereoisomers thereof: (I) In this formula, Y is N or CH, provided that when at least one of U, V, or W is N, Y may be CH only; U is N or CR &lt; 4 &gt;; V and W are independently N or CH provided that, U, V or W only one of which is at the same time may be nitrogen, R 4 is hydrogen, methyl or halogen; R &lt; 1 &gt; is phenyl or heteroaryl and is optionally substituted with halogen, lower alkyl or lower alkoxy; R is hydrogen or lower alkyl; R 2 is hydrogen, lower alkyl, lower alkoxy, CF 3 or S-lower alkyl; R 3 and R 3 ' are, independently of each other, hydrogen, lower alkyl or lower alkoxy, R 3 and R 3 ' together form a C 3 -5 -cycloalkyl-, tetrahydrofuran- or oxetane-ring. It has been found that the compound of formula I is an allosteric modulator of metabotropic glutamate receptor subtype 5 (mGluR5).
