abstract | The present invention describes a shortened method for preparing a solution comprising substantially purified purified capsular polysaccharide from cell streptococcus pneumoniae lysate culture. The clarified S. pneumoniae lysate is ultrafiltered and dyed and then the pH is adjusted to less than 4.5, preferably about 3.5, and more than 98% of the protein in the solution is precipitated without seriously affecting the polysaccharide yield . It is then subjected to ultrafiltration and diafiltration, acidification to below pH 4.5, filtration using activated charcoal, and precipitation of more than 90% of the residual protein without significant effect on polysaccharide yield. Non-limiting examples of S. pneumoniae serotypes that can be purified using the abbreviated process of the present invention include Type 1, 4, 5, 6A, 6B, 7F, 9V, 14, 18C 19A type, 19F type, and 23F type. In one embodiment, streptococcus pneumoniae cells are lysed using sodium deoxycholate (DOC), while in another embodiment the solubilizer is dissolved in a non-animal derived solubilizing agent such as N-lauryl sarcosine sodium (NLS )to be. |
