Vector for gene therapy |
abstract |
The present invention relates to a retrovirus comprising a transcriptional unit consisting of a transcriptional regulatory sequence and a gene encoding a polypeptide having endoribonuclease activity specific to a single-stranded RNA arranged in a form capable of controlling expression by the sequence. As a vector, there is provided a retroviral vector, wherein the unit is arranged such that the transcription direction of the mRNA of the unit is reverse to the transcription direction of the RNA genome of the retroviral vector. By using the vector of the said structure, the virus supernatant which shows high gene transduction efficiency can be prepared. The retroviral vector of the present invention is useful for the treatment and prevention of cancer or viral infections. |
