abstract | The present invention relates to compositions and methods for modulating α-secretase and / or improving cognition. The invention further relates to the treatment of diseases through improved / enhanced cognitive abilities, and increased sAPP production in individuals with disease, in particular Alzheimer's disease patients. Macrocyclic lactones (ie bryostatins and neristatins) are preferred compounds for use with the compositions of the present invention. The present invention also provides a method of increasing the production of non-amyloidogenic soluble APP, including activating PKC by administering an effective amount of protein kinase C (PKC) activator (s). |
