abstract | A method and kit for use in detecting any one of multiple or specific mutations, in particular multiple chromosomal abnormalities resulting from microdeletion, in a single assay. Polymerase chain reaction (PCR) is performed to amplify eukaryotic genomic DNA using a plurality of primer oligonucleotide pairs, with one primer in each pair having a detectable label attached to its 5 'end. Multiple primer pairs target DNA fragments of those different chromosomes that exhibit potential chromosomal abnormalities, and one pair targets control genes. The amplified PCR product is purified to obtain single-stranded DNA with a detectable label from which the single-stranded DNA comprises a DNA oligonucleotide probe having a nucleotide sequence complementary to one strand of nucleotide sequence of each fragment. Hybridize to points on each containing microarray. This microarray is imaged for the presence of a label at each point thereof, first comparing the results of this image with an image of a point specific to the control gene, and then comparing the results obtained from imaging of normal DNA to the target of interest.
