abstract The present invention relates to a medium composition comprising neuropeptide Y (Neuropeptide Y; NPY) effective for undifferentiated proliferation and maintenance of pluripotent stem cells, and to a method for culturing undifferentiated pluripotent stem cells using the same. By using neuropeptide Y as an undifferentiated pluripotent stem cell culture medium composition, it is possible to effectively proliferate and culture undifferentiated pluripotent stem cells, and in particular, under conditions that do not use feeder cell derived factors or animal serum. It was confirmed that long-term continuous passage of undifferentiated pluripotent stem cells could be performed with similar performance to that of co-culture or conditioned medium. Therefore, the present invention can be usefully used to improve the culturing process of pluripotent stem cells in undifferentiated state and ultimately develop a mass culture system capable of securing clinically applicable pluripotent stem cell resources. In addition, the present invention relates to a medium composition for dedifferentiation comprising neuropeptide Y (Neuropeptide Y; NPY) and a method for dedifferentiation using the same. More specifically, neuropeptide Y may be added to a medium composition for dedifferentiation induction. In this case, the efficiency of dedifferentiation could be significantly improved, and the induced pluripotent stem cells prepared by the above method were confirmed to have normal characteristics as pluripotent stem cells. Therefore, the present invention can be usefully used to develop stem cell cell therapeutics by improving dedifferentiation-inducing culture process and contributing to the development of clinically applicable induced pluripotent stem cell production technology.
