abstract | In addition to overexpression of related enzymes that mediate the synthetic pathway of GDP-fucose, the present invention is directed to recombinant E. coli recombinantly overexpressing glucose-6-phosphate-dehydrogenase or / and guanosine-inosine kinase. On how to produce GDP-fucose Using a recombinant E. coli co-expressing glucose-6-phosphate dehydrogenase (G6PDH) and ManB, ManC, Gmd and WcaG, a representative NADPH reproductive enzyme present in E. coli, pH-stat fed-batch culture produced up to 235.2 ± 3.3 mg / L of GDP-fucose, a 21% improvement over recombinant E. coli without G6PDH. In addition, using recombinant E. coli additionally expressing guanosine-inosine kinase (Gsk), GDP-fucose of up to 305.5 ± 5.3 mg / L was produced in a fed-batch culture of pH-stat method. This is a 58% improvement over recombinant E. coli without further expression of Gsk. |
