abstract | The present invention relates to a method for in vitro phosphorylation of Staphylococcus aureus TRAP protein. The present invention newly discovered that TRAP protein is a kinase that phosphorylates itself, and for the first time disclosed a method for phosphorylating purified TRAP protein. That is, the TRAP protein receives phosphate ions from ATP in the presence of iron, copper and zinc ions, demonstrating that the TRAP protein is phosphorylated. The present invention provides a method for phosphorylating TRAP protein in vitro and a method for effectively screening phosphorylation inhibitors of TRAP protein from various novel compounds and natural products using the method. Since phosphorylation is essential for the function of TRAP proteins, the development of inhibitors that inhibit this phosphorylation process provides the basis for the development of new antibiotics for staphylococci.
