The present invention provides an inexpensive and sensitive system and method for detecting analytes present in a medium. The system includes diffractive enhancing elements such as functionalized microspheres that have been modified to bind to the target analyte. In addition, the system includes a polymer film in which certain specific patterns of analyte-specific receptors can include a metal coating printed thereon. When attaching a target analyte directly or with a diffractive enhancing element to a selected region of the polymer film, diffraction of transmitted and / or reflected light occurs through the physical dimensions and defined exact placement of the analyte.
