titleOfInvention Modified Immune Numericin Vaccine Composition
abstract The present invention relates to modified numericin polypeptides which retain the immunogenic properties of numericin while being so small or reduced that no hemolytic activity is detected as compared to native numericin. The present invention also relates to methods of generating new numericin variants with these desired properties. In addition, the present invention provides an immunogenic composition useful as a pharmaceutical composition comprising a vaccine wherein the non-toxic, modified pneumericin stimulates prophylactic immunity against Streptococcus pneumoniae. The vaccine may be a composition in which a modified numericin is conjugated to a bacterial polysaccharide or may be included in a weakened viral vector. In addition, the present invention also uses a non-toxic, modified pneumolysin toxin to stimulate antibodies against Streptococcus pneumoniae in the treated subject and then isolates and delivers them to the secondary subject, thereby eventually streptococci in the secondary subject. It also relates to methods of providing Cous pneumoniae prophylaxis.
