The present invention relates to a method for preparing a protein having factor VII coagulation precursor activity under protein-independent conditions. Typically, this method comprises the steps of (1) inducing a highly productive, stable human cell line clone of Factor VII that lacks the B domain, and (2) acclimatizing the cells to grow in medium without plasma-derived protein. . More specifically, the method comprises transfecting a human cell line with a vector comprising a selectable marker as shown in the figure and a coding sequence of a protein having factor VIII coagulation precursor activity, selecting cells with a selection agent, and Isolating a clone that expresses high levels of a protein having factor VIII coagulation precursor activity.n n n n Factor VII, Coagulation Precursors, Cell Lines, Mass Production |
