abstract | The present invention relates to a purification process for separating target proteins by expressing the fusion partner and the target protein in a fused form, selectively adsorbing the matrix, and performing an effective cleavage reaction on the fusion partner and the target protein. According to the present invention, in a fusion protein comprising a protein of interest and a fusion partner, the fusion partner comprises an amino acid sequence cleaved by ubiquitin degrading enzyme at the C-terminus and the difference in the charge of the target protein and the isoelectric point of the fusion partner is different. Expressing a fusion protein in which the charge of the fusion partner is changed to be at least one, loading the fusion protein into the matrix, treating the fusion protein with ubiquitin degrading enzymes after being adsorbed on or separated from the matrix, and By recovering the protein, the protein of interest can be simply purified in high purity and yield. According to the present invention, it is possible to solve the complex purification process required for the production of recombinant protein, which can significantly improve the economics. |
