abstract | The present invention provides a method for identifying, isolating and producing htrB variants of pathogenic Gram-negative bacteria. The method mutated the htrB gene of a pathogenic Gram-negative bacterium to lack a functional htrB protein, thus lacking at least one secondary acyl chain contained in the wild-type pathogenic Gram-negative bacterium and substantially reduced compared to the wild-type strain. You get variants that show toxicity. In addition, the present invention relates to a method in which an htrB variant, an endotoxin isolated therefrom or an endotoxin isolated therefrom is conjugated with a carrier protein to immunize an individual against infection by a pathogenic Gram-negative bacterium by administering a prophylactically effective amount of a vaccine formulation. A method of using a vaccine formulation containing the same is provided. |
