A drug-highly enriched enteric-coated pharmaceutical composition comprising a core comprising an acid-sensitive drug in a low pH environment above pH 3, such as ddI, preferably comprises a methacrylic acid copolymer, a plasticizer, and an anti-adhesive agent. It is in the form of a beadlet having an enteric coating formed from an additional covering comprising. The beadlet has excellent resistance to disintegration at pH 3 or below but excellent drug properties at pH 4.5 and above. Also disclosed are methods of preparing the pharmaceutical compositions. |
