abstract | The present invention relates to nucleic acid molecules and expression vectors comprising a heat-induced promoter and host cells comprising at least one nucleic acid molecule of the present invention. More specifically, the present invention relates to kits and methods for producing one or more proteins using the nucleic acid molecules of the present invention, and various uses thereof. It is an object of the present invention to provide a promoter with a heat-inducible characteristic as selectively as possible, in particular to provide a promoter which is active in yeast and suitable for protein expression at high temperatures. An object of the present invention was accomplished by a nucleic acid molecule comprising a heat-induced promoter selected from the following nucleic acids: (a) a protein having trehalose-6-phosphate synthase activity; Nucleic acids having an array comprising the promoter sequence of the Hansenula polymorpha encoding; (b) a nucleic acid having the sequence set forth in SEQ ID NO: 1 ; (c) a nucleic acid having a sequence that exhibits at least 40% homology over a length of 300 bp with one of the sequences set forth in (a) or (b); (d) nucleic acids capable of hybridization with the complementary strand of one of the nucleic acids set forth in (a), (b) or (c); (e) derivatives derived from one of the nucleic acids set forth in (a), (b) or (c) obtained through substitution, addition, and / or deletion of one or more nucleotides ( derivative); (f) a fragment resulting from one of the nucleic acid molecules set forth in (a) to (e), having the function of a heat-induced promoter; (g) the combination of several nucleic acids set forth in (a) to (f), wherein the sequences of the nucleic acids may be different or the same; Or a nucleic acid molecule having a sequence complementary to one of the nucleic acids set forth in (a) to (g). |
