abstract The present invention relates to a method for producing a protein, wherein a protein refolded in a single step from a cell lysate including an inclusion body and a denaturant can be obtained to prepare an active protein in a short time, simply and economically. In order to provide a method for producing a protein, (A) the process of crushing a host cell containing a protein produced in the form of an inclusion body, and adding a denaturant to the crushed host cell to unfold the aggregated protein, ( B) adsorb the released protein to the solid matrix by introducing a cell lysate containing the denaturant and the released protein into the upper direction of a column in which a solid matrix having a higher density than the cell debris is suspended in an equilibrium state in the liquid. (C) adsorbing the adsorbed protein by introducing a buffer solution containing a denaturant into the column in an upward direction; Removing the cell lysate except from the column; (d) refolding the released protein by gradually reducing the concentration of the denaturant by supplying a buffer solution containing no denaturant to the column in an upward direction; and E) providing a method for producing a protein comprising the step of obtaining the refolded protein from the column.
