abstract The present invention provides an antibody comprising (a) a targeting sequence; (b) regulatory sequences: (c) exons; And (d) an unpaired splice-donor site. The present invention further relates to a method for producing a protein in vitro or in vivo, including homologous recombination of constituents in a cell as described above. Homologous recombinant cells are then maintained under conditions that allow transcription and translation to occur and obtain protein expression. In addition, the invention provides homologous recombinant cells comprising primary, secondary, or immortalized vertebrate cells, methods of making such cells, methods of generating fusion genes by homologous recombination, methods of altering gene expression in cells, and A method of making a protein in a cell using the constructs of the invention.
