abstract | The present invention relates to a search kit for searching for substances that exhibit an antibiotic effect by inhibiting the activity of aminoacyl-tRNA synthetase, and is easily and economically active against 20 aminoacyl-tRNA synthetases present in Escherichia coli. It is designed to search for inhibitors. Therefore, before performing in vitro assays, a method is required to be able to assay more economically, radioactive materials, and to rapidly assay activity inhibitors on tRNA syntheses and to confirm the effects on cells. This search kit solves this problem. In other words, aminoacyl-tRNA synthetase, which is an enzyme present in living organisms, is an enzyme that is essential for protein synthesis, and since there are 20 kinds of enzymes and high diversity, it is an option to search for active inhibitors. The range is wide and antibiotics have not been developed yet, so it is useful as a system for selecting antibiotics with new mechanisms. |
