abstract |
A) from about 40% to about 95% acetaminophen; b) a direct compression excipient comprising from about 1% to about 60% by weight of microcrystalline cellulose; And c) from about 0.01% to about 4.0% by weight of a pharmaceutically-acceptable lubricant. Acetaminophen and direct compression excipients are combined under shear mixing conditions sufficient to convert the acetaminophen and the direct compression excipient into homogeneous granules without collapse. In a preferred aspect of the invention, a lubricant is mixed with the acetaminophen and the direct compression excipient under high shear conditions. Also disclosed herein are methods of making direct compressed solid dosage forms and methods of treatment by the dosage forms disclosed herein. The method is particularly suitable for the preparation of direct compression dosage forms containing a high amount of acetaminophen (ie at least 80% by weight) based on the total weight of the tablet. |
