abstract |
By inserting DNA regulatory elements capable of promoting the expression of gene products normally expressed in cells, genes that are normally inactive at transcription can be activated in cell lines or microorganisms. This regulatory element is inserted to be expressively linked to a gene that is normally inactive. The insertion process is performed by creating a DNA construct containing fragments (targeting DNA) with DNA fragments of genes that are normally inactive by means of homologous recombination and DNA regulatory elements that induce transcription. This technique can also be used to modify the expression characteristics of endogenous genes of a given cell line or microorganism. |
