abstract The present invention relates to a novel succinic semialdehyde dehydrogenase (EC 1.2.1.24.) Isolated from bovine brain and a method for producing the same. The succinic semialdehyde dehydrogenase of the present invention is a dimer of about 110 kDa composed of two monomers each having a molecular weight of about 54 kDa, and has a very high substrate specificity for succinic semialdehyde, and an optimum pH of enzymatic activity is 9.0. Cysteine, lysine and arginine are involved in the activity. In addition, the succinic semialdehyde dehydrogenase of the present invention is treated with dapsone as an antiepileptic agent, diphenylhydantoin as an anticonvulsant and diazapam, and imipramine as an antidepressant / anti-anxiety agent. Activity is greatly inhibited. Accordingly, SSDH of the present invention may be applied to the development of inhibitors and activators of SSDH, and may be helpful in the treatment of various abnormalities caused by excessive accumulation of GHB and diseases caused when the body's level of GABA is abnormally low.
