abstract | In the present invention, the recombinant E. coli containing an inducible promoter using L-arabinose operon of Salmonella strains is cultured in a medium capable of high cell culture, and the expression is optimal at the optimal time considering cell growth and PTH expression. It provides a fed-batch batch fermentation method capable of inducing mass production of recombinant PTH. According to the fed-batch fermentation method of the present invention, by using the medium developed in the present invention and carrying out a fed-batch culture using the amount of dissolved oxygen as an indicator, the expression concentration minimum concentration obtained from the above experiments was determined by the concentration of glucose in the medium during the fed-batch culture. It was able to maintain the following, it can express a similar degree of PTH without expression inhibition compared to when using the glycerol as a carbon source. |
