abstract | The present invention relates to a novel gamma-aminobutyrate aminotransferase (EC 2.6.1.19.) Isolated from bovine brain. In the present invention, purified from bovine brains, GABA-T is a dimer of about 100,000 Daltons composed of the same two chains of monomers each having a molecular weight of about 50,000 Daltons, and 43% of the structure of GABA-T is α-helix (α- helix). In addition, as a result of examining specificity of several substrates, it was found that GABA-T performs an effective amino group transfer reaction on the amount of GABA in vivo. Thus, GABA-T was purified and biochemical and molecular biological studies of this enzyme were performed to compare the structure and amino acid sequence of GABA-T with porcine brain tissue or liver, and to explore the structure of the enzyme activity site. The present invention provides basic information on the development of inhibitors of GABA-T that can be raised to treat epilepsy. |
