abstract | The present invention provides a method for producing a heparosan compound in which the hexuronic acid residue is isomerized and a method for producing heparan sulfate, with improved C5-epimerization efficiency. Specifically, the present invention includes generating a heparosan compound in which a hexuronic acid residue is isomerized from a heparosan compound in the presence of a protein selected from the group consisting of (A) to (F) below. A method for producing a heparosan compound in which hexuronic acid residues are isomerized is provided: (A) a protein comprising the amino acid sequence of SEQ ID NO: 2; (B) a protein comprising an amino acid sequence having 90% or more homology with the amino acid sequence of SEQ ID NO: 2 and having D-glucuronyl C5-epimerase activity; (C) a protein having an amino acid sequence in which one or several amino acid residues are deleted, substituted, added or inserted in the amino acid sequence of SEQ ID NO: 2 and having D-glucuronyl C5-epimerase activity; (D) a protein comprising the amino acid sequence of SEQ ID NO: 5; (E) a protein comprising an amino acid sequence having 90% or more homology with the amino acid sequence of SEQ ID NO: 5 and having D-glucuronyl C5-epimerase activity; and (F) one or a number in the amino acid sequence of SEQ ID NO: 5 A protein comprising an amino acid sequence in which one amino acid residue is deleted, substituted, added or inserted, and having D-glucuronyl C5-epimerase activity. |
