abstract | A polypeptide having heparosan-glucuronic acid-5-epimerase activity is provided, and thus a means for producing a polysaccharide in which hexuronic acid residues are isomerized is provided. Heparosan-glucuronic acid, which is an isomerizing enzyme that screens African mimai cDNA library and acts on the glucuronic acid residue of N-acetylheparosan and / or the iduronic acid residue of fully desulfated / N-acetylated heparin A DNA encoding a polypeptide of -5-epimerase is obtained. The polypeptide encoded by this DNA is expressed in insect cells to obtain a polypeptide having heparosan-glucuronic acid-5-epimerase activity. By contacting this polypeptide with N-acetylheparosan or fully desulfated / N-acetylated heparin, a polysaccharide in which hexuronic acid residues are isomerized is obtained. |
