abstract Although it was thought that promotion of polysome formation in which a large number of ribosomes were attached to mRNA was very effective for highly efficient protein synthesis, the mechanism of the poly180 formation promotion function of p180 protein was not clear. The inventors of the present application identified SF3b4 as a protein that specifically interacts with the coiled-coil domain, which is a responsible region for the polysome formation promoting function of the p180 protein, and a protein that promotes endoplasmic reticulum (ER) localization of mRNA. Localization of mRNA on the endoplasmic reticulum in cells that express new proteins and that highly express both proteins that promote mRNA endoplasmic reticulum (ER) localization (eg, SF3b4 protein) and p180 protein As a result, it was revealed that the secretory function of cultured cells can be increased. In addition, by inserting a specific nucleotide sequence into the expression plasmid, it is possible to localize the SF3b4 protein, which has a protein expression enhancing effect, on the endoplasmic reticulum membrane, and to shift the weight of mRNA in the polysome.
