abstract | A non-natural protein containing at least one ester bond in the polypeptide main chain is synthesized using an in vivo translation system in ribosomes. (A) an aminoacyl tRNA synthetase capable of activating α-hydroxy acid, (b) a suppressor tRNA capable of binding to the α-hydroxy acid in the presence of the aminoacyl tRNA synthetase, and (c) nonsense at a desired position. A gene encoding a desired protein having undergone mutation or frameshift mutation is expressed in a cell or cell extract in the presence of the α-hydroxy acid. |
