The present invention provides a polynucleotide that has a high RNA interference effect on a target gene and has a very low risk of causing RNA interference for a gene that is not related to the target gene. From the base sequence of the target gene of RNA interference, a sequence site according to the following rules (a) to (d) is searched, and a polynucleotide that generates RNAi is designed and synthesized based on the search result. (A) The 3 'terminal base is adenine, thymine or uracil. (B) The 5 ′ terminal base is guanine or cytosine. (C) In the sequence of 7 bases at the 3 'end, one or more bases selected from the group consisting of adenine, thymine and uracil are rich.
