abstract Whether or not a specific amino acid residue of a protein is modified or the type of modifying group is easily determined. A reagent that labels the side chain of a specific amino acid residue of the protein to be analyzed is allowed to act. At this time, if the side chain of the specific amino acid residue is in an unmodified state, the side chain is labeled (S101). Then, the protein labeled with the side chain of a specific amino acid residue is specifically cleaved at a predetermined position (S102). Next, the obtained peptide fragment is subjected to mass spectrometry (S103). Based on the result obtained with respect to the molecular weight of the fragment, the modification state of the side chain of a specific amino acid residue in the protein to be analyzed is analyzed (S104).
