abstract | The present invention provides a single-chain antibody that retains its specific binding activity with an antigen and a labeled single-chain antibody in which a labeling substance is bound to the single-chain antibody. Specifically, the labeled single chain antibody of the present invention can be produced by binding a labeling substance to the linker portion of the single chain antibody. The antibody is produced using a cell-free protein synthesis system derived from wheat germ, and the antibody is produced in a low reducing state in which the intramolecular disulfide bond is retained. Further, a method for producing a solid-phase immobilized single chain antibody by binding the antibody to a solid phase via a labeling substance and a method for analyzing an antigen-antibody reaction using the solid-phase immobilized single chain antibody are performed. |
