abstract | The present invention relates to a plasmid in which a chromosome partitioning protein mukB gene on a bacterial chromosome is integrated with a fusion gene obtained by binding a fluorescent marker gene downstream of the mukB gene, a drug resistance gene, and a temperature-sensitive mutant gene encoding a replication initiation protein. A test substance is brought into contact with the host cell in which the homologous recombination gene has been replaced, and the presence or absence of the formation of a fluorescent focus is observed. Thus, a MukFEB complex composed of MukF, MukE and MukB proteins responsible for the main function of chromosome segregation is obtained. The purpose of the present invention is to provide a simple and rapid method for searching for a novel and useful antibacterial agent, which screens for substances that inhibit body formation. It is extremely useful as an antibacterial agent based on a novel mechanism of action. |
