abstract | PURPOSE:To carry out fractionation into 1,3-diglyceride lipase and 2- monoglyceride lipase, by subjecting a lipase obtained by cultivating Candida.cylindraceae to treatment using column chromatography. CONSTITUTION:An ion exchange gel, such as DEAE-Sephadex(R) or SP- Sephadex(R), or gel for gel filtration, such as Sephadex(R) G-150 or G-100, or both are used as a carrier, treated with MacIlvaine buffer solution (0.01mol. citric acid and 0.02mol. disodium phosphate; pH 3.8), etc., filled in a column having 1-10cm diameter, 50-200cm length, etc., and used for column chromatography. A lipase obtained by cultivating Candida.cylindraceae, such as lipase OF, is treated by the above-mentioned column chromatography and fractionated into 1,3-diglyceride lipase and 2-monoglyceride lipase. |
