abstract PURPOSE:To secure the reproducibility and stability of a developed color, by impregnating filter paper with nucleotide phosphorylase, xanthine oxidase, peroxidase, 2,7-diaminofluolein and o-tolidine. CONSTITUTION:Freshness testing paper is prepared by impregnating filter paper with nucleotide phosphorylase, xanthine oxidase, peroxidase, 2,7-diaminofluolein and o-tolidine. As the filter paper, filter paper for a chromatograph is pref. used. For example, the freshness testing paper is prepared by a method wherein water is added to 5ml of a phosphate buffer solution with pH of about 6-8, about 0.01-0.05ml of 2,7-diaminofluolein, about 0.01-0.05ml of o-tolidine, about 5-15 enzymatic unit of xanthine oxidase, about 10-30 enzymatic unit of nucleo tide phosphorylase, about 0.5-2g of sucrose (enzyme stabilizer) and 0.2ml of an about 0.5-2% aqueous mercaptoethanol (enzyme oxidation inhibitor) solution to adjust the whole to 15ml and this enzyne-dye solution is absorbed by the filter paper and the impregnated one is freeze-dried under vacuum.
