abstract | PURPOSE:To obtain a modified superoxide dismutase having high activity and excellent thermal stability, by linking a manganese type superoxide dismutase produced by a bacterium belonging to the genus Bacillus to polysaccharides. CONSTITUTION:Polysaccharides, such as pectin, carrageenan, alginic acid, agar, mannan, xanthan gum, dextran, pullulan, inulin, gum arabic, chitosan, carboxymethyl cellulose, methyl cellulose and aminoethyl cellulose, having >=2,000 average molecular weight are activated with a suitable reagent or binder and then active parts of the above-mentioned polysaccharides are linked to amino groups of superoxide dismutase in a neutral or weak alkaline solution at ambient temperature to carry out modification of the superoxide dismutase. The excessive active groups in the active polysaccharides are subsequently reacted with a compound having amino group to carry out post-treatment and the residual unreacted reagents, etc., are removed by ultrafiltration, etc., to afford the aimed modified superoxide dismutase. |
