abstract | PURPOSE:To selectively obtain the titled compound under mild condition in high yield, by reacting a compound such as N-ethoxycarbonyl-3-bromo-2-(2- oxypropyl)pyridine with a strong base and reacting the product in the presence of an acid catalyst. CONSTITUTION:The objective beta-substituted enamines of formula III [e.g. N- ethoxycarbonyl-5-(2-oxypropyl)-1,2,3,4-tetrahydropyridine] is produced by reacting a compound of formula I (R<1> and R<2> are H or 1-3C alkyl; R<3> is 1-10C alkyl or phenyl; X is halogen) with a strong base such as 1,8-diazabicyclo[5,4,0]-7- undecene to form a carbon-carbon bond at the beta-position of N of the compound of formula I and reacting the resultant compound of formula II in the presence of a catalytic amount of an acid (e.g. d,l-camphorsulfonic acid). The compound of formula III is a useful synthetic intermediate for various drugs, perfumes and natural alkaloid compounds. |
