abstract NEW MATERIAL:The polypeptide having an amino acid sequence of formula. USE:An antitumor agent. PREPARATION:For example, gene of human tumor necrosis factor (TNF) is chemically synthesized by selecting the amino acids constituting human TNF protein from a designated codon. The gene is linked with a proper vector and closed to obtain an expression-type plasmid. The plasmid is cut with a proper restriction enzyme to remove a specific region in the human TNF gene and the gene is repaired by using an oligonucleotide having another base sequence to obtain a recombinant plasmid containing a DNA region coding a polypeptide having methionine group bonded to an amino terminal. The plasmid is inserted into a microorganism and the transformed microorganism is cultured to obtain the polypeptide having the amino acid sequence of formula.
