titleOfInvention Immunoassay method using amphoteric surfactant
abstract PURPOSE:To enhance the sensitivity of immunoassay in immunohistocyoto chemical measurement by conjugating the serum to be inspected, antibody and labeling antibody and leaning the conjugate thereof with a cleaning liquid added with an ionic amphoteric surfactant. CONSTITUTION:The cell membrane of, for example, the single-layered Langerhans island cells of the rat pancreas is utilized as the antigen. This membrane is fixed and is cleaned by a physiological salt soln. of a phosphoric acid buffer. The dilute liquid of the serum to be inspected is then added and conjugated to and with this membrane and the conjugate is cleaned with the cleaning liquid added with the ionic amphoteric surfactant. The dilute liquid of the labeling antibody is added and conjugated to and with conjugate and the conjugate is cleaned with the above-mentioned cleaning liquid. Dodecyl-M- betaine (lauryl dimethyl betaine aminoacetate), etc., are used as the ionic amphoteric surfactant. The labeling antibody is thereafter measured by a fluorescent method. Since the conjugate is cleaned by the cleaning liquid added with the ionic amphoteric surfactant, the specific staining of the labeling antibody is easily discriminated and the specimen is measured with high sensitivity.
