abstract | PURPOSE:To obtain DNA to code bovine adrenal adrenodoxin reductase, by selecting the DNA from a cDNA bank prepared from bovine adrenal by immunochemical method using an antibody to adrenodoxin reductase. CONSTITUTION:A cDNA library comprising lambda gt11 is prepared from mRNA of bovine adrenal and screened by using lambda gt11 immuno-screening kit to give one positive clone. Phage DNA is prepared from the positive clone, cleft with ECoRI, cDNA is recovered, inserted into ECoRI site of plasmid pCU 19, introduced into Escherichia coli to give transformant JM 103 (pADR). The transformant is cultivated to prepare plasmid DNA, which is treated with a restriction enzyme to give the aimed cDNA of pADR with 2.0kb. |
