abstract | PURPOSE: To obtain the titled substance useful as medicines, food additives, etc., on an industrial scale, advantageously, by adding excess alkaline agent to a solution of acidic salt of protamine to dissociate anion and precipitating the protamine by salting-out with an organic solvent or an alkaline agent. n CONSTITUTION: A solution of protamine acidic salt produced e.g. by extracting from spermatozoid nucleus of salmon, trout, walleye pollack, etc., is added with ≥5 times equivalent of an alkaline agent [e.g. NaOH, KOH, Ba(OH) 2 , etc.] (based on the anion bonded to protamine) to isolate protamine. The isolated protamine is precipitated by adding usually ≥2 times volume of an organic solvent (e.g. ethanol, methanol, acetone, etc.) (based on the protamine solution) or adding an alkaline agent to attain an alkali concentration of ≥10%, thereby effecting the salting-out of the protamine. n COPYRIGHT: (C)1988,JPO&Japio |
