abstract | PURPOSE: To measure a protein specific antigen derived from bacteria with high sensitivity, by using a peroxidase-labelled antibody represented by a specific formula as a measuring reagent. n CONSTITUTION: At first, a solid phase sensitized with an antibody is brought into contact with an antigen (a bacteria culture solution or a specimen itself such as sputum or laxative feces) to perform antigen-antibody reaction. Next, after the solid phase is washed, a labelled antibody represented by formula (I) is added to the solid phase to form an aggregate having a sandwich like structure of antibody-antigen-antibody. The solid phase is again washed and the activity of peroxidase bonded to the solid phase through the antigen is measured. The measurement of activity is performed by a method wherein trimethylbenzidine is added to perform reaction and 2M sulfuric acid is added to the reaction mixture as a reaction stopping agent and color development peculiar to the substrate reacted is utilized to measure absorbancy.
