titleOfInvention Plasmid having ability to decompose vanillic acid
abstract PURPOSE:To raise the productivity of an enzyme decomposing vanillic acid, by cleaving chromosome of Pseudomonas paucimobilis with a restriction enzyme and linking the formed fragment containing an enzyme gene capable of decomposing vanillic acid therein to a plasmid. CONSTITUTION:Cultivated microbial cells of Pseudomonas paucimobilis are treated to prepare a chromosomic DNA, which is cleaved with a restriction enzyme EcoRI to collect a fragment having enzymic gene capable of decomposing vanillic acid. The resultant fragment and a fragment obtained by cleaving a separately prepared plasmid pKT230 with the EcoRI are mixed and linked with a T4 ligase to afford the aimed plasmid having the ability to decompose vanillic acid. The resultant plasmid having the ability to decompose vanillic acid can be inserted into a host of Escherichia coli, bacterium of the genus Pseudomonas, etc., to provide a transformant.
