NEW MATERIAL:A monoclonal antibody reactive specifically with D monomer in plasmin decomposition product of human fibrinogen and D dimer in plasmin decomposition product of human fibrin but non-reactive with fibrinogen, X, Y and E. n USE: A reagent for clinical diagnosis of diffused intravascular coagulation syndrome (DIC) for quantitative determination of D dimer and (DD)E complex. n PREPARATION: For example, a mouse is immunized with a solution of D-dimer immunogen by peritoneal infusion. A spleen cell is collected from the mouse and subjected to cell fusion with a mouse myeloma cell to obtain a hybridoma. The hybridoma is cultured in a HAT medium, screened with an immunogen to separate a hybridoma capable of producing anti-D-dimer antibody, monoclonized by limiting dilution analysis and cultured to obtain the objective monoclonal antibody. n COPYRIGHT: (C)1988,JPO&Japio |
