titleOfInvention Production of synthetic intermediate for optically active carbacycline
abstract PURPOSE: To easily produce an optically active compound useful as a synthetic intermediate for carbacycline, at a low cost, by treating a specific racemic compound with a lipase, thereby effecting selective asymmetric hydrolysis. n CONSTITUTION: A racemic material which is a mixture of the compounds of formula I and formula II is treated with a lipase (e.g. lipase originated from Aspergillus niger) to effect the selective hydrolysis of only the acetoxy group of the compound of formula I and the conversion of the compound to the optically active compound of formula III to obtain a mixture of the optically active compound of formula III and the optically active compound of formula II. The reaction product is extracted with a solvent such as ether, the solvent is distilled out and the obtained compounds of formula II and formula III are separated from each other e.g. by column chromatography. The objective optically active compound of formula III which is an alcoholic compound of the compound of formula I useful as a synthetic intermediate for carbacycline can be produced by this process. n COPYRIGHT: (C)1988,JPO&Japio
