abstract | PURPOSE:To obtain industrially carnitine having high optical purity at a low cost and in high yield, by reacting carnitine amide with a specific enzyme. CONSTITUTION:A microorganism which produces an enzyme capable of converting carnitine amide into carnitine is separated from the natural world to provided a strain of the genus Pseudomonas, CA32-C (FERM P-8911), which is subjected to shake culture at about 26 deg.C or 72hr in a culture medium containing glucose, meat extract, etc., and DL-carnitine amide hydrochloride, etc., to provided a culture medium containing an enzyme (amidase) having an activity by which carnitine amide is hydrolyzed to afford carnitine. Then, 0.1-30wt% carnitine amide (e.g. DL-carnitine amide hydrochloride) which is a substrate is added to the medium and reacted with the enzyme medium at 4-1 pH and 0.5-60 deg.C for 1-100hr. The resultant resultant liquid is treated with an ion exchange membrane, etc., to recover an optically active carnitine. |
