abstract PURPOSE: To measure an enzyme in body fluid simply and in high sensitivity, by using a composition for multi-layer analysis consisting of an upper layer containing a water-insoluble substrate, bonded to a coenzyme, of an object enzyme to be measured and a lower layer containing an action enzyme of the coenzyme and the substrate of the enzyme. n CONSTITUTION: An upper layer 2 contains a water-soluble or insolubilized substrate, bonded to a coenzyme such as NAD, NADH, NADP, ATP, etc., of an object enzyme to be measured. The object enzyme to be measured is amylase, collagenase, trypsin, elastase, etc., and the substrate of the enzyme is selected from starch, collagen, elastin and casein. An action enzyme such as glucose-6- phosphate dehydrogenase, tryptophanase, etc., of the coenzyme and the substrate of the action enzyme are added to a lower layer 4. Each layer is laminated to a substrate and a reverse development preventing layer 3 is laid between the upper layer and the lower layer to give a composition for multi-layer analysis. n COPYRIGHT: (C)1988,JPO&Japio
