abstract | PURPOSE: To obtain the titled enzyme having high titer at a low cost, by using as an enzyme source, a waste liquid of a process for producing wheat starch from wheat flour composed of gluten layer and embryo of wheat grain. n CONSTITUTION: A 3rd-grade wheat flour (usually called as C-flour) produced from hard or semi-hard wheat is used as a raw material and subjected to a hydration and kneading step, a step to wash the dough with water to separate starch milk and gluten, a step to separate starch milk and waste liquid and a drying step of starch milk. The waste liquid generated in the above wheat starch production process is purified e.g. by ultrafiltration, salting-out with ammonium sulfate, sodium chloride, etc., coagulation with a flocculant such as aluminum sulfate, precipitation with an organic solvent, concentration purification by distilling out water under reduced pressure and if necessary, gel-filtration with anion exchange resin column chromatography or batchwise purification process.
