abstract PURPOSE:To product CPD-choline in high efficiency, by using CMP and choline or phosphorylcholine as a substrate and contacting with a CPCT clone having increased activity of choline phosphate cytidyl transferase. CONSTITUTION:Whole DNA of Saccharomyces cerevisiae X2180-1 B is digested with a restriction enzyme SauBAI to obtain a DNA fragment of 5-15kb. A fragment produced by digesting vector YEp13 with BamHI is linked with the above DNA fragment using T4 ligase to obtain a recombinant DNA. A gene library of Saccharomyces cerevisiae is prepared by transforming E.coli K12 strain with the recombinant DNA. Saccharomyces cerevisiae D319-8A strain is transformed with said library to obtain CPCT clone. The cultured liquid, microbial cell or their treated product of the clone is made to contact with CMP (cytidine-5'-monophosphate) and choline or CMP and phosphorylcholine in a reaction liquid to obtain CDP-choline (cytidine diphosphocholine).
